چکیده

During the last decade, the application of pharmacokinetic and pharmacodynamic modeling techniques has become an increasingly important aspect of contemporary clinical psychopharmacology (1–5). These techniques have been applied during the process of development of new drug entities as well as for the improved understanding of the clinical actions of drugs that are already marketed. Techniques for the study of drug metabolism in vitro have advanced substantially during the last decade, and now are an integral component of preclinical drug development and the link to subsequent clinical studies of drug metabolism and disposition. Kinetic-dynamic modeling techniques have been combined with in vitro metabolism procedures and in vitro–in vivo mathematical scaling models to provide insight into the general problem of pharmacokinetic drug interactions in clinical psychopharmacology (6–9). This chapter reviews some advances in pharmacokinetics, pharmacodynamics, and drug metabolism, along with methodologic applications to selected problems in clinical psychopharmacology.

Effect of the CYP2C8 genotype on the pharmacokinetics and pharmacodynamics of repaglinide.

The pharmacokinetics of repaglinide shows pronounced interindividual variability, for which several reasons have been considered, including interactions with drugs inhibiting CYP2C8 and CYP2C8 genetic polymorphism. However, existing data on the role of genetic polymorphisms in repaglinide disposition are not fully consistent. We studied the effect of CYP2C8*3 on the pharmacokinetics and pharmac...

Pharmacokinetics and Pharmacodynamics of Gliclazide from Immediate and Modified Release Formulation Tablets in Rats

The objective of the study was to compare pharmacokinetic and pharmacodynamic parameters of gliclazide after administration of immediate (IR) and modified release (MR) tablets. The experiment included rats with both normoglyceamia and streptozocin (STZ)-induced hyperglyceamia. Several MR formulations were designed and in vitro drug release profile was assessed by a dissolution test. For the fur...

Comparative tolerability, pharmacodynamics, and pharmacokinetics of a metabolite of a quinolizinone hypnotic and zolpidem in healthy subjects.

The objectives of this double-blind, placebo-controlled study were to assess the single dose tolerability, pharmacodynamics, and pharmacokinetics of Ro 41-3290 (5, 10, and 30 mg) and zolpidem (10 mg) in three sequential groups of 10 healthy male subjects. Pharmacodynamic (tracking, attention, and memory test) and pharmacokinetic measurements were conducted over a period of 24 and 50 h, respecti...
